Azide-PEG3-FLAG
Azide-PEG 3-FLAG enables the FLAG-tag (DYKDDDDK) attachment to any terminal Alkyne-or Cyclooctyne (e.g. DBCO) -functionalized molecule via Cu (I) -catalyzed terminal Alkyne-Azide Click Chemistry (CuAAC) or Cu (I) -free strain-promoted Alkyne-Azide Click Chemistry (SPAAC), respectively. The resulting FLAG-tagged molecules can subsequently be de-tected by an anti-FLAG antibody that is either immobilized onto a matrix (for purification) or coupled with a fluorescent dye or reporter enzyme for direct or indirect detection, respectively. Molecule solu-bility and efficient FLAG-tag detection is ensured by the integrated PEG-linker. Azide-FLAG (without PEG-linker) has been successfully used for the detection of Azide-tagged glycoproteins in cell lysates (final concentration: 100 M) . This concentrations may serve as a starting point for individual assay set-up.
