RUNX1/ETO tetramerization-IN-1 is a potent small-molecule inhibitor that disrupts the NHR2-mediated tetramerization of the RUNX1/ETO oncoprotein (EC50 = 0.25 μM) . It restores downregulated gene expression, inhibits RUNX1/ETO-dependent leukemia cell proliferation in vitro, and demonstrates significant anti-tumor efficacy in mouse models of AML.
