GSK3494245 (DDD01305143) is a potent, selective, and orally bioavailable inhibitor targeting the chymotrypsin-like activity of the Leishmania donovani proteasome with an IC50 of 0.16 μM. It also inhibits the human 26S proteasome (IC50=13 μM) and demonstrates utility for researching kinetoplastid parasites in both in vitro and in vivo models.
