PD146176 (NSC-168807) is a potent 15-lipoxygenase (15-LO) inhibitor, demonstrating a Ki of 197 nM in enzymatic assays. In vivo studies using aged 3xTg-AD mice show it can ameliorate Alzheimer's-like pathology, including cognitive deficits and amyloidosis, by promoting autophagic clearance mechanisms.
