Celastrol
Celastrol (Tripterin), a bioactive compound from Thunder God Vine, is a potent proteasome inhibitor that preferentially targets the 20S chymotrypsin-like activity (IC50 = 2.5 μM) and inhibits the 26S proteasome in prostate cancer cells. It induces apoptosis, promotes accumulation of ubiquitinated proteins and proteasome substrates, suppresses AR expression, and demonstrates significant tumor growth inhibition in vivo.
