Product Specifications

Background

PHB2 (Prohibitin 2), also called Repressor of Estrogen Receptor Activity (REA), is a protein that in humans is encoded by the PHB2 gene. The International Radiation Hybrid Mapping Consortium mapped the PHB2 gene to chromosome 12. Montano et al. (1999) showed that REA enhanced the potency of a dominant-negative ER-alpha mutant and antiestrogens as suppressors of ER-alpha activity in Chinese hamster ovary cells. When coexpressed with wildtype ER-alpha or ER-beta (ESR2), REA suppressed activation of a reporter gene in a dose-dependent manner. REA had no effect on reporter activity in the absence of liganded ER, and it had no effect on the transcriptional activities of other hormone receptors. Mutation analysis showed that an N-terminal domain and a central domain of REA were required for its repressor activity.

Immunogen

A synthetic peptide corresponding to a sequence in the middle region of human REA, identical to the related rat and mouse sequences.

Clonality

Polyclonal

Tissue Specificity

Expressed on platelets and leukocytes and is primarily concentrated at the borders between endothelial cells. Isoform Long predominates in all tissues examined. Isoform Delta12 is detected only in trachea. Isoform Delta14-15 is only detected in lung. Isoform Delta14 is detected in all tissues examined with the strongest expression in heart. Isoform Delta15 is expressed in brain, testis, ovary, cell surface of platelets, human umbilical vein endothelial cells (HUVECs), Jurkat T-cell leukemia, human erythroleukemia (HEL) and U-937 histiocytic lymphoma cell lines (at protein level). .

IHC,ICC,WB

Purification

Immunogen affinity purified.

Concentration

Adding 0.2 ml of distilled water will yield a concentration of 500 μg/ml.

Form

Lyophilized

Reconstitution

Add 0.2ml of distilled water will yield a concentration of 500ug/ml.

Function

Acts as a mediator of transcriptional repression by nuclear hormone receptors via recruitment of histone deacetylases (By similarity). Functions as an estrogen receptor (ER)-selective coregulator that potentiates the inhibitory activities of antiestrogens and represses the activity of estrogens. Competes with NCOA1 for modulation of ER transcriptional activity. Probably involved in regulating mitochondrial respiration activity and in aging. .

Components

Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg Thimerosal, 0.05mg NaN3.
