It utilizes the red fluorescent label sulforhodamine 101 (Texas Red), which is linked to the target amino acid phenylalanine, and a chloromethyl ketone (CMK) reactive group, forming the fluorescent inhibitor probe, SR101-Phe-CMK. SR101-FLISP labels active chymotrypsin-like serine proteases in living cells.
