HLA-DQ monoclonal antibody (clone SPV-L3) is a mouse IgG2aκ antibody that specifically recognizes the human HLA-DQ molecule, a class II major histocompatibility complex (MHC) protein. HLA-DQ is a heterodimer composed of an alpha (DQA1) and beta (DQB1) chain, both anchored in the membrane of antigen-presenting cells (APCs) such as B cells, dendritic cells, and macrophages. SPV-L3 was raised against a T4-positive cytotoxic T lymphocyte clone (HG-38) and binds a monomorphic determinant of HLA-DQ. It is widely used in flow cytometry, immunocytochemistry, immunofluorescence, and immunohistochemistry, with validated reactivity in human and porcine samples. Importantly, SPV-L3 has been shown to block cytotoxic activity of T4-positive CTL clones, making it useful in functional assays and immune regulation studies. This antibody is often used in transplantation research, autoimmune disease studies, and immunopeptidomics, where HLA-DQ plays a role in antigen presentation and immune recognition.
