RANKL ("Receptor Activator of NF-κB Ligand") is a protein of the tumor necrosis factor (TNF) family discovered in 1997 that is significantly involved in the regulation of bone remodeling. The protein is also known as TRANCE (TNF-related activation-induced cytokine), CD254, OPGL, and ODF. RANKL is secreted by, among others, osteoblasts (cells that build bone substance) and binds to the "matching" receptor RANK of monocytic osteoclast progenitor cells, inducing their differentiation into osteoclasts (cells that degrade bone substance again) . The RANK/RANKL system is a biochemical regulatory circuit that causes bone resorption to remain in healthy balance with bone formation, which is a prerequisite for the dynamic architecture of the bone system.
