Background

PPM1H (Protein Phosphatase, Mg2+/Mn2+ Dependent 1H) is a serine/threonine phosphatase that modulates key signaling pathways in cellular homeostasis. It acts as a negative regulator of LRRK2 signaling by dephosphorylating Rab GTPases, which are critical for vesicle trafficking and lysosomal function. PPM1H has been identified as a gatekeeper of bone morphogenetic protein (BMP) signaling, preventing excessive activation through targeted dephosphorylation. It also serves as an independent prognostic biomarker in non-small cell lung cancer. In neuroscience, PPM1H’s regulation of Rab proteins and BMP pathways positions it as a potential modulator of neuroinflammation and synaptic integrity. Its role in maintaining vesicular transport and signaling balance makes it a candidate for therapeutic exploration in neurodegenerative diseases such as Parkinson’s.
