Prolyl 3-hydroxylase that catalyzes the post-translational formation of 3-hydroxyproline on collagens (PubMed:18487197) . Contributes to proline 3-hydroxylation of collagen COL4A1 and COL1A1 in tendons, the eye sclera and in the eye lens capsule (By similarity) . Has high activity with the type IV collagen COL4A1, and lower activity with COL1A1 (PubMed:18487197) . Catalyzes hydroxylation of the first Pro in Gly-Pro-Hyp sequences where Hyp is 4-hydroxyproline (PubMed:18487197) . Has no activity on substrates that lack 4-hydroxyproline in the third position (PubMed:18487197) .

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Leprecan Like Protein 1 (LEPREL1) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Leprecan Like Protein 1 (LEPREL1) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Leprecan Like Protein 1 (LEPREL1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Leprecan Like Protein 1 (LEPREL1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
