2-oxoglutarate dehydrogenase (E1) component of the 2-oxoglutarate dehydrogenase complex, which mediates the decarboxylation of alpha-ketoglutarate (PubMed:24495017) . The 2-oxoglutarate dehydrogenase complex catalyzes the overall conversion of 2-oxoglutarate to succinyl-CoA and CO (2) (PubMed:24495017) . The 2-oxoglutarate dehydrogenase complex is mainly active in the mitochondrion (PubMed:29211711) . A fraction of the 2-oxoglutarate dehydrogenase complex also localizes in the nucleus and is required for lysine succinylation of histones: associates with KAT2A on chromatin and provides succinyl-CoA to histone succinyltransferase KAT2A (PubMed:29211711) .

Assay Principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Oxoglutarate Dehydrogenase (OGDH) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Oxoglutarate Dehydrogenase (OGDH) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Oxoglutarate Dehydrogenase (OGDH), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Oxoglutarate Dehydrogenase (OGDH) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
