Target

Calcium channel that mediates the release of Ca (2+) from the sarcoplasmic reticulum into the cytoplasm and thereby plays a key role in triggering muscle contraction following depolarization of T-tubules (PubMed:11741831, PubMed:16163667) . Repeated very high-level exercise increases the open probability of the channel and leads to Ca (2+) leaking into the cytoplasm (PubMed:18268335) . Can also mediate the release of Ca (2+) from intracellular stores in neurons, and may thereby promote prolonged Ca (2+) signaling in the brain. Required for normal embryonic development of muscle fibers and skeletal muscle. Required for normal heart morphogenesis, skin development and ossification during embryogenesis (By similarity) .

Assay Principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Ryanodine Receptor 1, Skeletal (RYR1) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Ryanodine Receptor 1, Skeletal (RYR1) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Ryanodine Receptor 1, Skeletal (RYR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Ryanodine Receptor 1, Skeletal (RYR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
