Thermolysin-like specificity, but is almost confined on acting on polypeptides of up to 30 amino acids (PubMed:15283675, PubMed:8168535) . Biologically important in the destruction of opioid peptides such as Met- and Leu-enkephalins by cleavage of a Gly-Phe bond (PubMed:17101991) . Able to cleave angiotensin-1, angiotensin-2 and angiotensin 1-9 (PubMed:15283675) . Involved in the degradation of atrial natriuretic factor (ANF) (PubMed:2531377, PubMed:2972276) . Displays UV-inducible elastase activity toward skin preelastic and elastic fibers (PubMed:20876573) .

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Neprilysin (CD10) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Neprilysin (CD10) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Neprilysin (CD10), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Neprilysin (CD10) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
