Its primary physiological function is unclear. May play a role in neuronal development and synaptic plasticity. May be required for neuronal myelin sheath maintenance. May promote myelin homeostasis through acting as an agonist for ADGRG6 receptor. May play a role in iron uptake and iron homeostasis. Soluble oligomers are toxic to cultured neuroblastoma cells and induce apoptosis (in vitro) (By similarity) . Association with GPC1 (via its heparan sulfate chains) targets PRNP to lipid rafts. Also provides Cu (2+) or ZN (2+) for the ascorbate-mediated GPC1 deaminase degradation of its heparan sulfate side chains (By similarity) .

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Prion Protein (PRNP) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Prion Protein (PRNP) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Prion Protein (PRNP), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Prion Protein (PRNP) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
