Intra-assay Precision (Precision within an assay) : 3 samples with low, middle and high level Cyclin Dependent Kinase 4 (CDK4) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays) : 3 samples with low, middle and high level Cyclin Dependent Kinase 4 (CDK4) were tested on 3 different plates, 8 replicates in each plate. Ser/Thr-kinase component of cyclin D-CDK4 (DC) complexes that phosphorylate and inhibit members of the retinoblastoma (RB) protein family including RB1 and regulate the cell-cycle during G (1) /S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complexes and the subsequent transcription of E2F target genes which are responsible for the progression through the G (1) phase. Hypophosphorylates RB1 in early G (1) phase. Cyclin D-CDK4 complexes are major integrators of various mitogenenic and antimitogenic signals. Also phosphorylates SMAD3 in a cell-cycle-dependent manner and represses its transcriptional activity. Component of the ternary complex, cyclin D/CDK4/CDKN1B, required for nuclear translocation and activity of the cyclin D-CDK4 complex.

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cyclin Dependent Kinase 4 (CDK4) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Cyclin Dependent Kinase 4 (CDK4) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cyclin Dependent Kinase 4 (CDK4), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Cyclin Dependent Kinase 4 (CDK4) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
