Intra-assay Precision (Precision within an assay) : 3 samples with low, middle and high level Chemokine C-X3-C-Motif Ligand 1 (CX3CL1) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays) : 3 samples with low, middle and high level Chemokine C-X3-C-Motif Ligand 1 (CX3CL1) were tested on 3 different plates, 8 replicates in each plate. Chemokine (C-X3-C motif) ligand 1; Cx3c; CX3C membrane-anchored chemokine; C-X3-C motif chemokine 1; fractalkine; neurotactin; Scyd1; small inducible cytokine subfamily D, 1; small-inducible cytokine D1.

Acts as a ligand for both CX3CR1 and integrins. Binds to CX3CR1 and to integrins ITGAV:ITGB3 and ITGA4:ITGB1. Can activate integrins in both a CX3CR1-dependent and CX3CR1-independent manner. In the presence of CX3CR1, activates integrins by binding to the classical ligand-binding site (site 1) in integrins. In the absence of CX3CR1, binds to a second site (site 2) in integrins which is distinct from site 1 and enhances the binding of other integrin ligands to site 1. The soluble form is chemotactic for T-cells and monocytes, but not for neutrophils. The membrane-bound form promotes adhesion of those leukocytes to endothelial cells. May play a role in regulating leukocyte adhesion and migration processes at the endothelium.

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Chemokine C-X3-C-Motif Ligand 1 (CX3CL1) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Chemokine C-X3-C-Motif Ligand 1 (CX3CL1) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Chemokine C-X3-C-Motif Ligand 1 (CX3CL1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Chemokine C-X3-C-Motif Ligand 1 (CX3CL1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
