Target
C8 is a constituent of the membrane attack complex. C8 binds to the C5B-7 complex, forming the C5B-8 complex. C5-B8 binds C9 and acts as a catalyst in the polymerization of C9. The gamma subunit seems to be able to bind retinol.

Assay Principle
The microplate provided in this kit has been pre-coated with an antibody specific to Complement Component 8g (C8g) . Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Complement Component 8g (C8g) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Complement Component 8g (C8g) level in the sample or standard.
