Target
Transcription factor that specifically recognizes and binds E-box sequences (5'-CANNTG-3') . Efficient DNA-binding requires dimerization with itself or with another MiT/TFE family member such as TFEB or MITF. In association with TFEB, activates the expression of CD40L in T-cells, thereby playing a role in T-cell-dependent antibody responses in activated CD4 (+) T-cells and thymus-dependent humoral immunity. Specifically recognizes the MUE3 box, a subset of E-boxes, present in the immunoglobulin enhancer. It also binds very well to a USF/MLTF site.

Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Transcription Factor Binding To IGHM Enhancer 3 (TFE3) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Transcription Factor Binding To IGHM Enhancer 3 (TFE3) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Transcription Factor Binding To IGHM Enhancer 3 (TFE3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Transcription Factor Binding To IGHM Enhancer 3 (TFE3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
