Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1D gives rise to L-type calcium currents. Long-lasting (L-type) calcium channels belong to the 'high-voltage activated' (HVA) group. They are blocked by dihydropyridines (DHP), phenylalkylamines, benzothiazepines, and by omega-agatoxin-IIIA (omega-Aga-IIIA) . They are however insensitive to omega-conotoxin-GVIA (omega-CTx-GVIA) and omega-agatoxin-IVA (omega-Aga-IVA) .

Assay Principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcium Channel, Voltage Dependent, L-Type, Alpha 1D Subunit (CACNa1D) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcium Channel, Voltage Dependent, L-Type, Alpha 1D Subunit (CACNa1D) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcium Channel, Voltage Dependent, L-Type, Alpha 1D Subunit (CACNa1D), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Calcium Channel, Voltage Dependent, L-Type, Alpha 1D Subunit (CACNa1D) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
