Required to maintain individual mitotic chromosomes dispersed in the cytoplasm following nuclear envelope disassembly (PubMed:27362226) . Associates with the surface of the mitotic chromosome, the perichromosomal layer, and covers a substantial fraction of the chromosome surface (PubMed:27362226) . Prevents chromosomes from collapsing into a single chromatin mass by forming a steric and electrostatic charge barrier: the protein has a high net electrical charge and acts as a surfactant, dispersing chromosomes and enabling independent chromosome motility (PubMed:27362226) . Binds DNA, with a preference for supercoiled DNA and AT-rich DNA (By similarity) . Does not contribute to the internal structure of mitotic chromosomes (PubMed:26949251) . May play a role in chromatin organization (PubMed:26949251) . It is however unclear whether it plays a direct role in chromatin organization or whether it is an indirect consequence of its function in maintaining mitotic chromosomes dispersed.

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Ki-67 Protein (Ki-67) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Ki-67 Protein (Ki-67) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Ki-67 Protein (Ki-67), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Ki-67 Protein (Ki-67) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
