Target

Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca (2+) /calmodulin-binding and autophosphorylation, and is involved in sarcoplasmic reticulum Ca (2+) transport in skeletal muscle and may function in dendritic spine and synapse formation and neuronal plasticity. In slow-twitch muscles, is involved in regulation of sarcoplasmic reticulum (SR) Ca (2+) transport and in fast-twitch muscle participates in the control of Ca (2+) release from the SR through phosphorylation of the ryanodine receptor-coupling factor triadin. In neurons, may participate in the promotion of dendritic spine and synapse formation and maintenance of synaptic plasticity which enables long-term potentiation (LTP) and hippocampus-dependent learning (By similarity) .

Assay Principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcium/Calmodulin Dependent Protein Kinase II Gamma (CAMK2g) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcium/Calmodulin Dependent Protein Kinase II Gamma (CAMK2g) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcium/Calmodulin Dependent Protein Kinase II Gamma (CAMK2g), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Calcium/Calmodulin Dependent Protein Kinase II Gamma (CAMK2g) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
