Target
Mitochondrial outer membrane GTPase that mediates mitochondrial clustering and fusion (PubMed:11181170, PubMed:11950885, PubMed:28114303) . Mitochondria are highly dynamic organelles, and their morphology is determined by the equilibrium between mitochondrial fusion and fission events (PubMed:28114303) . Overexpression induces the formation of mitochondrial networks (PubMed:28114303) . Membrane clustering requires GTPase activity and may involve a major rearrangement of the coiled coil domains (Probable) . Plays a central role in mitochondrial metabolism and may be associated with obesity and/or apoptosis processes (By similarity) . Plays an important role in the regulation of vascular smooth muscle cell proliferation (By similarity) . Involved in the clearance of damaged mitochondria via selective autophagy (mitophagy) (PubMed:23620051) . Is required for PRKN recruitment to dysfunctional mitochondria (PubMed:23620051) . Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mitofusin 2 (MFN2) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Mitofusin 2 (MFN2) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mitofusin 2 (MFN2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Mitofusin 2 (MFN2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
