Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.

Assay Principle
Aviva Systems Biology ALB ELISA Kit (Horse) (OKCA00357) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Horse ALB antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated ALB and incubated. The ALB found in the sample or standards competes with the biotinylated ALB for limited binding sites on the immobilized anti-Horse ALB antibody. Excess unbound biotinylated ALB and sample or standard ALB is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated ALB captured in the well and inversely proportional to the amount of ALB which was contained in the sample or standard.
