Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.

Assay Principle
Aviva Systems Biology ALB ELISA Kit (Rabbit) (OKCA00226) is based on a competitive enzyme immuno assay technique. The microtiter well-plate in this kit has been pre-coated with Rabbit ALB and blocked. Standards or samples are added to the plate wells along with a horseradish peroxidase (HRP) conjugated antibody specific to ALB and incubated. ALB immobilized in the wells competes with the liquid phase ALB contained in the samples or standards for binding with the HRP-Conjugated ALB Antibody. Wells are washed removing unbound ALB, unbound HRP-Conjugated ALB Antibody or ALB/HRP-Conjugated ALB Antibody complex. HRP-Conjugated ALB Antibody bound to ALB immobilized in the well remains. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of HRP-Conjugated ALB Antibody captured in the well and inversely proportional to the amount of ALB which was contained in the sample or standard.
