Midkine (MK) is a member of a family of neurotrophic and developmentally-regulated heparin-binding molecules that currently consists of midkine, pleiotrophin/PTN and avian RI-HB. This gene is mapped to 3q24. Across species, MK shows 87% amino acid identity between the human and mouse proteins. Between family members, human MK is approximately 50% identical to human PTN.

Aviva's rat Midkine ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Midkine has been precoated onto 96-well plates. Standards (Expression system for standard: E.coli, Immunogen sequence: V21-D140) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Midkine is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the rat Midkine amount of sample captured in plate.
