Target
This PROS1 gene encodes a vitamin K-dependent plasma protein that functions as a cofactor for the anticoagulant protease, activated protein C (APC) to inhibit blood coagulation. This gene is mapped to 3q11.1. It is found in plasma in both a free, functionally active form and also in an inactive form complexed with C4bbinding protein. Mutations in this gene result in autosomal dominant hereditary thrombophilia. An inactive pseudogene of this locus is located at an adjacent region on chromosome 3. Alternative splicing results in multiple transcript variants encoding different isoforms that may undergo similar processing to generate mature protein.

Assay Principle
Aviva's human PROS1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PROS1 has been precoated onto 96-well plates. Standards (Expression system for standard: NS0, Immunogen sequence: A42-S676) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PROS1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Fibronectin amount of sample captured in plate.
