PCSK9 is the 9th member of the proprotein convertase family of proteins that activate other proteins.PCSK9 has medical importance because it acts in lipoprotein homeostasis. PCSK9 is ubiquitously expressed in many tissues and cell types. PCSK9 binds to the receptor for low-density lipoprotein particles (LDL), which typically transport 3,000 to 6,000 fat molecules (including cholesterol) per particle, within extracellular fluid. The LDL receptor (LDLR), on liver and other cell membranes, binds and initiates ingestion of LDL-particles from extracellular fluid into cells, thus reducing LDL particle concentrations.

Aviva's rat PCSK9 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PCSK9 has been precoated onto 96-well plates. Standards (Expression system for standard: NS0, Immunogen sequence: S156-Q694） and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PCSK9 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the rat PCSK9 amount of sample captured in plate.
