Klotho (KL) is a glycosylated protein that plays an important role in the regulation of phosphate and calcium homeostasis. This gene encodes a type-I membrane protein that is related to beta-glucosidases. Reduced production of this protein has been observed in patients with chronic renal failure (CRF), and this may be one of the factors underlying the degenerative processes (e.g., arteriosclerosis, osteoporosis, and skin atrophy) seen in CRF. Also, mutations within this protein have been associated with ageing and bone loss and alcohol consumption.

Aviva's human Klotho ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Klotho has been precoated onto 96-well plates. Standards (NS0, E34-S981) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Klotho is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Klotho amount of sample captured in plate.
