Target
NOV (nephroblastoma overexpressed), also known as CCN3, is a matricellular protein that in humans is encoded by the NOV gene. The protein encoded by this gene is a small secreted cysteine-rich protein and a member of the CCN family of regulatory proteins. This gene is mapped to 8q24.12. NOV is a potentially useful marker for the diagnosis of adrenal gland diseases, malignant adrenocortical tumors, multiple sclerosis and so on. Moreover, reduced expression of NOV in ACTs may play an important role in the process of childhood ACT tumorigenesis. Though studying, it identified Nov as a regulator of human hematopoietic stem or progenitor cells.

Assay Principle
Aviva's mouse Ccn3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for Ccn3 has been precoated onto 96-well plates. Standards (Expression system for standard: NS0, Immunogen sequence: Q22-I354) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ccn3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the mouse Ccn3 amount of sample captured in plate.
