Target
FGF2 has been implicated in a multitude of physiologic and pathologic processes, including limb development, angiogenesis, wound healing, and tumor growth. Human FGF2 shares 96% and 97% amino acid sequence homology with mouse and rat respectively. FGF2 belongs to the fibroblast growth factor (FGF) family. Fibroblast growth factors (FGFs) exhibit widespread mitogenic and neurotrophic activities. Nine members of the family are currently known, and FGF-1 and FGF-2 are present in relatively high levels in CNS. FGF-2 is expressed by at low levels in many tissues and cell types and reaches high concentrations in brain and pituitary.

Assay Principle
Aviva's human FGF2 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for FGF2 has been precoated onto 96-well plates. Standards (E.coli, P143-S288) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for FGF2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human FGF2 amount of sample captured in plate.
