Target
Cathepsin S, also known as CTSS, is a protein that in humans is encoded by the CTSS gene. A member of the peptidase C1 family, CTSS gene is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. By fluorescence in situ hybridization (FISH), this gene is mapped to 1q21.3. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. The bond-specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N.

Assay Principle
Aviva's human Cathepsin S ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Cathepsin S has been precoated onto 96-well plates. Standards (NS0, Q17-I331 & S109-I331) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Cathepsin S is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Cathepsin S amount of sample captured in plate.
