Product Specifications

Background

Alpha-1-antitrypsin?or?α1-antitrypsin? (A1AT, ?A1A, or?AAT) is a?protein?belonging to the?serpin?superfamily. It is encoded in humans by the?SERPINA1?gene.?The?gene?is located on the long arm of the fourteenth?chromosome? (14q32.1) .The protein encoded by this gene is secreted and is a serine protease inhibitor whose targets include elastase, plasmin, thrombin, trypsin, chymotrypsin, and plasminogen activator. Defects in this gene can cause emphysema or liver disease. Several transcript variants encoding the same protein have been found for this gene.Like all?serine protease inhibitors, A1AT has a characteristic?secondary structure?of?beta sheets?and?alpha helices.?Mutations?in these areas can lead to non-functional proteins that can?polymerise?and accumulate in the?liver (infantile hepatic cirrhosis) .

Cross Reactivity

There is no detectable cross-reactivity with other relevant proteins.

Immunogen

Expression system for standard: NS0; Immunogen sequence: E25-K418

Tissue Specificity

Ubiquitous. Expressed in leukocytes and plasma.

Applications

ELISA

Field of Research

Intracellular, Kinases, Metabolism, Metabolism Processes, Pathways and Processes, Protease Inhibitors, Proteolysis/Ubiquitin, Response To Hypoxia, Serum Proteins

Sample Type

Cell culture supernatants, serum, plasma (heparin, EDTA) and urine.

Detection Range

1.56 ng/ml - 100 ng/ml

Sensitivity

<10 pg/ml

Function

Inhibitor of serine proteases. Its primary target is elastase, but it also has a moderate affinity for plasmin and thrombin. Irreversibly inhibits trypsin, chymotrypsin and plasminogen activator. The aberrant form inhibits insulin-induced NO synthesis in platelets, decreases coagulation time and has proteolytic activity against insulin and plasmin.

Storage Conditions

Store at 4°C for 6 months, at -20°C for 12 months. Avoid multiple freeze-thaw cycles (Ships with gel ice, can store for up to 3 days in room temperature. Freeze upon receiving.)
