Neuroligin 1 (NLGN1) is a postsynaptic cell adhesion molecule critical for the formation and maturation of excitatory synapses. As a member of the neuroligin family, NLGN1 interacts with presynaptic neurexins to establish trans-synaptic connections that regulate synaptic specificity and plasticity. In the adult brain, NLGN1 is essential for long-term potentiation (LTP), synaptic remodeling, and memory formation, particularly within the hippocampus and amygdala. Dysregulation of NLGN1 expression or function has been linked to impaired synaptic transmission, cognitive deficits, and neurodevelopmental disorders. In neurodegenerative diseases, such as Alzheimer’s disease, synaptic loss precedes neuronal death. Altered NLGN1 signaling may contribute to early synaptic dysfunction, making it a potential biomarker and therapeutic target. NLGN1 also participates in activity-dependent synaptic scaling, suggesting a role in maintaining excitatory-inhibitory balance under pathological conditions. Understanding NLGN1’s role in synaptic maintenance and plasticity provides insight into the molecular mechanisms underlying cognitive decline and offers avenues for synapse-targeted therapies in neurodegeneration.

2 µg/ml of SMC-463 was sufficient for detection of Neuroligin-1 in 20 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.
