Background

LRP4 (Low-Density Lipoprotein Receptor-Related Protein 4) is a transmembrane receptor essential for neuromuscular junction formation and synaptic signaling. It acts as a co-receptor for agrin and MuSK, facilitating synapse development and maintenance. In the central nervous system, LRP4 influences astrocyte-neuron communication and synaptic plasticity. Mutations or autoantibodies targeting LRP4 are linked to myasthenia gravis and may contribute to cognitive dysfunction. Emerging evidence suggests LRP4 plays a role in neurodegenerative diseases by modulating synaptic integrity and glial function. Its involvement in both peripheral and central synaptic regulation positions LRP4 as a promising target for therapeutic intervention in neuromuscular and neurodegenerative disorders.
